Injectables for human and veterinary administration
When drugs are be administered by parenteral route, solubilization plays a more important role than in oral administration, as with the latter, a soild dosage form is equally acceptable. Therefore the pyrogen-free types,PVP(Povidone) K17 are recommended for parenterals. The molecular weight of this product is low enough enough(mw 9000) to allow repaid renal elimination without storage.
In some countries in Europe, like Germany and Austria, only such low-molecular polyvidone types with a K-value of up to 18 are approved for injection.
PVP(Povidone) K17 is nowadays widely used in antibiotic and sulfonamide formulations. Table 1 shows, how the solubilization of rifampicin depends directly on the povidone concentration.
Table 2 shows the composition of a commercially available oxytetracycline preparation for injection, that containsPVP(Povidone) K17 as a solubilizing agent.
Table 2: Composition of a commercially available oxytetracycline ampoule
A positive side-effect of the use of PVP(Povidone) K17 is a reduction in the local toxicity of oxytetracycline.(from Volker Buhler-- polyvinylpyrrolidone for the pharmaceutical industry)
